RESEARCH PROJECT IN PROTECTIVE AND ANTI-INFLAMMATORY PROPERTIES OF SIgA

In this research project, we focus on the role of secretory immunoglobulin A (SIgA) in gut homeostasis. First, using an in vitro model, we have shown by laser scanning confocal microscopy that commensal bacteria such as Lactobacilli or Bifidobacteria interact with SIgA in a Fab-independent manner through carbohydrates moieties present on the secretory component, indicating a new role of SIgA in sensing the microbiota. Furthermore, using monolayers derived from a human intestinal epithelial cell line, we have demonstrated that the pathogenic bacteria Shigella flexneri associated with SIgA has reduced impact on the monolayers. Among others, the presence of SIgA limits inflammatory responses such as the activation of the NF-B pathway. All together, these results indicate that SIgA represent one of the pivotal elements in the fragile intestinal balance.
