Description :

The proteomics unit is a service at the CNP open for all research groups at the University of Lausanne and is an extension of the Proteomic Analysis Facility (PAF), UNIL. Its mission is to develop novel technologies for the analysis of protein samples and to facilitate the identification of surrogate markers.

Services consist in :

  • Anaylsis of protein samples by one- & two-dimensional
    gel electrophoresis.

  • Various protein stains (Coomassie blue, Silver-,
    Sypro Ruby- and Phospho-stains).

  • Western blots, and analysis of immunoprecipates
    for associated proteins.

  • Computer assisted identification of protein differences
    with Image Master software.

  • Differential gel electrophoresis of infrared-labeled protein samples for spot identification.

  • Serial protein labeling in redox proteomics for the identification of oxidized proteins.

  • Dysregulation of the ubiquitin proteasome system in Alzheimer's disease

Various protein stains (Coomassie blue, Silver-, Sypro Ruby- and Phospho-stains)

At the same time, the research group of Dr. B. Riederer is working on neurodegeneration and investigates metabolic changes in Alzheimer’s disease and Schizophrenia by collaborating with different groups at the CNP. One aspect is protein oxidation and its influence on protein ubiquitination and degradation. We, and others have demonstrated that protein ubiquitination in Alzheimer’s disease is dysfunctioning and may contribute towards protein aggregation and neuronal death. In Schizophrenia the anti-oxidation system of glutathione is not functioning correctly due a genetic variation of a key enzyme and thus may underly the importance of a deficient anti-oxidation defense mechanism in this multifactorial disorder.
